abstract PURPOSE: To enable the easy and accurate determination of methotrexate useful as a chemotherapeutic agent for leukemia, in the blood fluid, by adding β- cyclodextrin to a solution of dihydrofolic acid, thereby stabilizing said solution. n CONSTITUTION: A solution of dihydrofolic acid (abbreviated as FAH 2 ) is stabilized by adding β-cyclodextrin thereto. In the use of methotrexate, it is necessary to determine the concentration of the compound in the body fluid continuously for preventing the side effects. Although an enzymatic process using an enzyme, dihydrofolate reductase (abbreviated as DHFR) is suitable for the clinic test for the determination of methotrexate because of the simplicity in the operation, it has a problem that the FAH 2 solution used as the substrate of DHFR is unstable. The defect of the enzymatic process can be overcome by stabilizing the FAH 2 solution with β-cyclodextrin. n COPYRIGHT: (C)1984,JPO&Japio
