abstract | PURPOSE: To prepare high-purity malto-oligosaccharide useful as a substrate for the analysis of pharmaceuticals, foods, amylase, etc., easily, by contacting immobilized yeast cells with a sugar solution containing oligosaccharide. n CONSTITUTION: Yeast belonging to Saccharomyces genus, e.g. Saccharomyces cerevisiae, etc. is cultured aerobically in a nutrient medium containing maltose and/or maltotriose as a carbon source, and the yeast cells containing a substance having α-glucosidase activity produced and accumulated in the cell are immobilized to obtain immobilized yeast cells. Malto-oligosaccharides having a polymerization degree equal to or higher than maltotetraose and essentially free from oligosaccharide having molecular weight equal to or lower than maltotriose is prepared by contacting the immobilized yeast cells with a sugar solution containing oligosaccharides, thereby assimilating the low-molecular weight oligosaccharide having molecular weight equal to or lower than maltotriose. n COPYRIGHT: (C)1984,JPO&Japio |
