titleOfInvention Method and composition for determination of beta-hydroxybutyric acid
abstract PURPOSE: To determine β-hydroxybutyric acid easily and rapidly, by reacting β-hydroxybutyric acid with NAD in the presence of a dehydrogenase, reducing a tetrazolium salt with the produced NADH, and determining the amount of the produced formazan by colorimetry. n CONSTITUTION: β-Hydroxybutyric acid in a specimen is oxidized to acetoacetic acid with β-hydroxybutyric acid dehydrogenase in the presence of nicotinamide adenine dinucleotide (NAD). The tetrazolium salt added to the system {e.g. 3,3'- (3, 3'-dimethoxy-4, 4'-biphenylene)-bis[2-( p-nitrophenyl )-5-phenyl-2H-tetrazolium]} is reduced by the reducing NAD (NADH) produced by the above oxidation reaction, by the aid of an electron transfer material (e.g. methyl-1-methoxy-5-methylphenazium sulfate), and the produced colored formazan is determined by colorimetry to determine the β-hydroxybutyric acid (cf. reaction formula). n COPYRIGHT: (C)1984,JPO&Japio
