abstract | PURPOSE: To provide a thrombolytic agent by using a specific plasminogen activator as an active component. n CONSTITUTION: The objective thromobolytic agent contains a plasminogen activator having the following characteristics: exhibits one protein band having an apparent molecular weight of 55,000±1,000 by the electrophoresis using sodium dodecyl sulfate-polyacrylamide gel; the apparent molecular weight is not influenced by the presence of a reducing agent; it has immonological properties which are not adsorbed by antiurokinase IgG-sephalose affinity chromatography; exhibits hydrolytic activity to the group I , and not to the group II. The above active component can be prepared by (1) extracting the human kidney, etc. in the presence of ammonium thiocyanate, (2) acidifying the extract to obtain precipitate, (3) extracting the precipitate with a buffer solution, and (4) purifying the extract by passing through a column containing an ion exchange material. The compound is extremely effective as a remedy for acute and chronic thrombosis, without causing side effects such as internal hemorrhage, anaphylactic shock, etc. n COPYRIGHT: (C)1984,JPO&Japio |
