abstract PURPOSE:A complex plasmid is synthesized from a plasmid of gram-positive Streptococcus faecalis and a plasmid vector of gram-negative Escherichia coli. CONSTITUTION:The objective complex plasmid includes the tetracycline-resistant gene area Tc originating from plasmid pAM-1 and the replication starting area OripAM1 originating from plasmid pAM1 in Streptococcus faecalis, further Kanamycin-resistant gene area Km originating from plasmid vector pACYC177 and the replication starting area Ori177 originating from the plasmid vector pACYC177 in Escherichia coli. Further, there is only one recognition cleavage site by the restriction enzyme BalI in the tetracycline-resistant gene area and recognition cleavage sites by the restriction enzymes XhoI and ClaI exist in the Kanamycin-resistant gene area, respectively.
