titleOfInvention Method and device for analyzing homovanilic acid and vanilylmandelic acid
abstract PURPOSE:To analyze homovanilic acid and vanilylmandelic acid with high accuracy by separating both acids from a soln. to be analyzed using a moving phase contg. N(C4H9)4, producing each dimer in the presence of of H2O2 with peroxidase, and measuring fluorescent light intensity of each dimer. CONSTITUTION:A moving phase contg. N(C4H9)4 is fed from a vessel 7 together with a sample from a sample introducing part to a reverse phase chromatographic column 4 to separate a vanilylmandelic acid (VMA) and homovanilic acid (HVA). The eluate is fed to a column 6 is mixed with a phosphoric acid buffer soln. contg. peroxidase fed from a vessel 7A and a phosphoric acid buffer soln. contg. H2O2 fed from a vessel 7B and reacted to form each dimer of the 2 acids. They are fed to a mixing coil 9 together with a mixed soln. of boric acid-sodium carbonate-NaOH of pH11-12 fed from a vessel 7C. Since the produced dimers exhibit fluorescent light at lambdaEx 315nm and lambdaEm 425nm, respectively, said lights are detected with a detector 10 and indicated with a recorder 11. Said acids are separated and determined thus exactly in a short time.
