abstract | PURPOSE:To increase the sensitivity of the electrode by fixing aldehyde dehydrogenase together with ADH and NAD on a collector such as carbon. CONSTITUTION:The mixture composed of the powder of carbon (graphite) as the collector, which serves also as a carrier for fixing enzyme and coenzyme, and of the powder of fluorocarbon resin as a binder is formed into a pellet by pressure forming, whereby a base body of electrode is prepared. On this base body of electrode, the water solution containing 100mg/ml of nicotinamide adenine dinucleotide (NAD) is developed at the rate of 40mul/cm<2> and dried. Next, the water solution containing dehydrogenase such as alcohol dehydrogenase (ADH) and aldehyde dehydrogenase in 50mg/ml each is developed at the rate of 40mul/ cm<2> and dried. Thereafter cross-linking reaction is caused by the action of glutaric aldehyde as a fixing reagent, washing is water is conducted, and thus the enzyme electrode A is obtained. This constitution enables removal of acetaldehyde which becomes an impediment factor and, therefore, a current increase value about twice as large as the one of a conventional electrode B is obtained. |
