abstract | PURPOSE: To increase sensitivity and to measure accurately and quickly in the determination of bile acids, by utilizing a combination of the final detecting system of measurement of bile acids using steroid dehydrogenase and an enzymic cycling reaction. n CONSTITUTION: When bile acids (e.g., glycollic acid, etc.) in an organism sample are converted into keto bile acids by 3α-hydroxysteroid dehydrogenase, the conversion of NAD + (or NADP + ) into NADH (or NADPH) is measured by using an enzymic cycling reaction (fig.). For example, in the fig., using an NAD cycling wherein A; oxaloacetic acid, A'; malic acid, Ea; malic dehydrogenase, B; ethyl alcohol, B'; acetaldehyde, Eb; glucose-6-phosphoric dehydrogenase, an increase in the amount of NADH is obtained by the absorbance at 340nm to determine the concentration of bile acids. n COPYRIGHT: (C)1982,JPO&Japio |
