titleOfInvention Production of (+)-5alpha-dihydropregnenolone
abstract PURPOSE: The reaction of a dodecahydrophenanthrene-2(1H)-one derivative with methyl lithium is followed by reaction of the product with a methyl halide in the presence of a metal amide and cyclization to give the titled compound in high yield through a shortened process. n CONSTITUTION: The reaction of a compound of formula I with methyl lithium is conducted in an organic solvent such as tetrahydrofuran to give an acetylenic alcohol of formula II. The product is made to react with a methyl halide such as methyl iodide in the presence of a metal amide to form a compound of formula III. The cyclization of the compound is effected with a strong acid in a solvent such as a mixture of trifluoroacetic anhydride-trifluoroacetic acid to give (+)-5α-dihydropregnenolone of formula IV. n EFFECT: In the process, the stereoselectivity is significantly kept to give the objective compound. n COPYRIGHT: (C)1982,JPO&Japio
