abstract PURPOSE:To prevent the occurrence of distrubance due to an inpurities and to enable to perform a micromeasurement, by a method wherein, after vincamin in an organism component is separated and refined, alkali hydrolysis is conducted and alcohol is further added to measure fluorescence. CONSTITUTION:After a collected sample is prepared into pH4-9 in prior to extraction, it is extracted with an organic solvent. In order to remove a disturbing substance such as vitamines, aminos, coloring matters, vincamin and other foreign elements are separated through separation and refinement using columm chromatography. An alkali hydrolysis is conducted on the obtained vincamin eluate after a solvent is removed by dirtillation. If heated to 50-70 deg.C thereupon, a steadier hydrolysis can be conducted in a short time. After the solution is cooled to a room temperature, alcohol such as methyl alcohol, ethyl alcohol is added to measure fluorescence. The addition of alcohol permits strengthening of the fluorescence and also enables to perform a measurement of 0.05mug/ml vincamin.
