titleOfInvention Method for improving immunity determination
abstract PURPOSE:To determine the activity of bound beta-galactosidase immune component in enzyme immuno assay by using a reaction medium containing at least one compound selected from among hydrophilic alcohols and ketones. CONSTITUTION:0.2ml Of 0.1M phosphoric acid buffer solution of pH 6.8 (containing 1mM MgCl2; 0.1M NaCl; 0.1% cattle serum albumin; 0.1% NaN3), 10mg/ ml nitrophenyl beta-D galactoside (buffer solution) and a reaction medium or a reference medium are mixed. The mixture is subjected to reaction for 50min in a water bath 37-45 deg.C. At the end of the reaction the reaction is interrupted by adding 2.5ml of 0.1M glycine-sodium carbonate buffer solution of pH 10.4 to the mixture and the light absorbance of the mixture is measured at 420nm to determine the enzyme activity (relative activity).
