titleOfInvention Preparation of 7alpha-acylthio-4-en-3-oxosteroid
abstract PURPOSE: To prepare the titled compound useful as an antialdosteroic diuretic which is an extremely effective remedy, in high yield, by adding a thiocarboxylic acid to 4,6-diene-3-oxosteroid in a specific solvent. n CONSTITUTION: The titled compound is obtained by adding a thiocarboxylic acid (e.g. thioacetic acid) to a 4,6-diene-3-oxosteroid (e.g. 17-hydroxy-3-oxo-17α-pregna- 4,6-diene-21-carboxylic acid γ-lactone, etc.) in a solvent containing N-methyl-2- pyrrolidone or N,N-dimethylacetamide. The reaction may be carried out if necessary, in the presence of a catalytic amount of an acid stronger than the thiocarboxylic acid. n EFFECT: The production of by-product is essentially completely suppressed and the purification efficiency is improved, which result in remarkable economical effects. Stable even by reacting for a long time. n COPYRIGHT: (C)1981,JPO&Japio
