Preparation of immunoglobulin with high content of monomer |
abstract |
PURPOSE: To obtain the titled compound useful for preventing and remedying various infectious diseases, by fractionating immunoglobulin-containing material in the presence of a specific basic compound with preventing the formation of aggregate with side effects. n CONSTITUTION: Immunoglobulin is collected from an immunoglobulin-containing material (e.g., serum or plasma) by fractionation. In the operation, a water-soluble solution of a basic nitrogen-containing organic compound (e.g., arginine) having a dissociation constant pKb not more than 7 is added to the immunoglobulin-containing material during the process of fractionation (especially, initial process), to give immunoglobulin with high content of monomer. The addition of the basic compound prevents the immunoglobulin monomer occurring during fractionation process from agglomerating into a dimer or polymer. These agglomerates activate a complement component, consequently various side effects are brought about. n COPYRIGHT: (C)1981,JPO&Japio |
