titleOfInvention | Preparation of peroxidase |
abstract | PURPOSE: To use a peroxidase as a labeling enzyme in immunoassays, e.g. clinical diagnosis, by cultivating a microorganism, belonging to the Myrothecium verrucaria, and capable of producing the peroxidase, and collecting the peroxidase from the resultant culture. n CONSTITUTION: A microorganism belonging to Myrothecium verrucaria, e.g. Myrothecium verrucaria IFO 6113 or IFO 6133, is cultivated. The spore, hypha or the preliminary culture obtained by cultivating the microorganism is inoculated into a liquid or solid culture medium. The cultivation is carried out in a liquid culture medium containing a carbon source, e.g. glucose or molasses, a nitrogen source, e.g. peptone or urea, and another minor nutrient source at about pH4W11 and a cultivation temperature of about 15W37°C by the spinner culture method with aeration or stationary culture method. In the case of a solid culture medium, the cultivation is carried out in bran, chaff, etc. for about 3W10 days. The aimed substance is collected from the culture fluid by the conventional method. n COPYRIGHT: (C)1981,JPO&Japio |
