titleOfInvention Preparation of optically active n-mercaptoacyl-imino acid
abstract PURPOSE: An optically active acid is used as a reagent for separating diastereoisomers and protecting agent to prepare title compound useful as hypotensor from a propionic acid derivative and L-proline with industrial advantage. n CONSTITUTION: For example, an optically active acid as 1-menthoxyacetic acid is made to react with racemic 3-mercapto-2-methylpropionic acid to give a diastereoisomer mixture of the compound of formula I. After each isomer is isolated, one of them is made to react with proline or its salt to form the compound of formula II or its salt. The product is treated with a diluted acid and the 1-menthoxyacetyl group is removed to give the objective compound of formula III. The removed optically active acid can be circulated. The 2(S)-isomer of the compound of formula I is readily led to the 2(S)-isomer of the compound of formula III with higher pharmaceutical activity by reaction with L-proline and deacylation. n COPYRIGHT: (C)1980,JPO&Japio
