NEW MATERIAL:7-Lysyl-amino-4-methylcoumarin derivative of formula (R is H, glutamyl, leucyl, isoleucyl-glutamyl, phenylalanyl-glutamyl, etc.), its acid addition salt, its hydrate, and its amino-protected derivative at the N<alpha>- or N<epsilon>-position. EXAMPLE:7-(N<alpha>-t-Butyloxycarbonyl-L-glutamyl-L-lysyl)-amino-4-met hylcoumarin. USE:Fluorescent substrate for the determination of enzyme activity, and its synthetic intermediate. It is hydrolyzed by enzymes such as trypsin, plasmin, etc. PROCESS:The compound of the formula is prepared by (1) reacting 7-amino-4- methylcoumarin with a protected lysine in the presence of a condensing agent, and (2) condensing the reaction product with a derivative of glutamic acid, leucine, etc. before or after removing the protecting group. |
