abstract | PURPOSE: To calculate aceto-acetic acid in blood with a high sensitivity simply by making aceto-acetic acid in blood into an azo compound by reaction on p-nitropheny diazonium fluoborate and converting this azo compound to a hydrazo compound and comparing color. n CONSTITUTION: Albuminous substances are eliminated from the blood serum of a tested person, and aceto-acetic acid is made into an azo compound by reaction on p-nitrophenyl diazonium fluoborate in citric acid buffer liquid of pH 4.5. Next, phosphorous acid is added to decompose an excess diazonium salt and convert the azo compound to a hydrazo compound, and this solution has the photo absorptivity measured by a wave length of 390nm, and the measured value is compared with a standard calculation line, thereby calculating aceto-acetic acid in blood.
