abstract |
PROBLEM TO BE SOLVED: To calibrate / standardize a flow cytometry device using synthetic polymer particles or beads having physical properties that provide advantages when used in the device. SOLUTION: Calibrated or standard synthetic particles have a predetermined volume and refractive index value, the device produces a light scattering related signal, a) for the particles to be tested, calibrated or standard synthetic particles. Passing through an incident beam of light; b) detecting first and second measurable signals from a calibration or standard composite particle passing through the incident beam of light; c) detecting the first and second signals. Measuring the intensity and generating a signal intensity pair, d) converting the signal intensity pair into volume and refractive index values, Applying a standardized signal gain to the flow cytometer; ande) measuring (i) volume and refractive index and (ii) Calibrating the flow cytometer for a count of analyzable particles having a signal within the appropriate volume and refractive index range. |
