Provided is a cleanliness test reagent capable of detecting even a small amount of dirt with high sensitivity and accurately evaluating cleanliness, and a cleanliness test method using the reagent. A dirt collection carrier is brought into contact with an inspection location, The soiled carrier is then contacted with sterile water or a reagent for extracting microbial intracellular components, and the resulting contact solution is added with (1) pyruvate orthophosphate dikinase, A cleanliness test reagent containing phosphoenolpyruvate, pyrophosphate, luciferin, luciferase and a metal salt; or (2) ADP from ADP to A A cleanliness test reagent to which at least one selected from the group consisting of an enzyme catalyzing the reaction for generating TP and an RNase is added, the amount of luminescence generated is measured, and the cleanliness test is performed.
