abstract (57) [Problem] To provide a method for producing a protein using a cell-free protein synthesis system, which can easily synthesize a sufficient amount of protein from DNA obtained by PCR. A method for producing a protein in a cell-free protein synthesis system containing a cell-free extract, comprising the steps of: An NA sequence, a ribosome binding DNA sequence, a DNA sequence encoding a target protein, and a terminator DNA sequence or a mutant terminator DNA sequence; A method for producing a protein using a template DNA having a total DNA sequence of 50% or more of the total DNA sequence.
