abstract | (57) [Problem] To provide a method for synthesizing a sugar chain using an immobilized glycosyltransferase. A method for producing a sugar chain, comprising the following steps. (1) A substance having a sugar residue bonded to a side chain of a water-soluble polymer via a linker containing a bond that can be selectively cleaved is used as a primer, and the primer is immobilized in the coexistence of a sugar nucleotide. (2) transferring the sugar residue of the sugar nucleotide from the sugar nucleotide to the sugar residue of the primer by contacting the sugar nucleotide with the glycosyltransferase; and (2) repeating step (1) once or twice or more. Extending a sugar chain by transferring a plurality of sugar residues, (3) If necessary, a step of removing by-produced nucleotides or unreacted sugar nucleotides, and (4) repeating steps (1) to (3) a plurality of times, and then transferring a plurality of sugar residues. A step of releasing the extended sugar chain by cleaving a selectively cleavable bond contained in the linker from the primer having an extended sugar chain, and a method for producing a glycosphingolipid. |
