abstract (57) [Problem] To provide a method for determining a base sequence of DNA using RNA polymerase, which can determine the base sequence without using a labeled terminator. SOLUTION: A nucleic acid transcription product obtained by reacting ribonucleoside 5 triphosphates and 3 deoxyribonucleoside 5 triphosphate in the presence of an RNA polymerase and a DNA fragment containing a promoter sequence for the RNA polymerase is separated. And a method for determining the nucleotide sequence of a DNA for reading the sequence of a nucleic acid from the obtained separated fraction, wherein the initiator for the nucleic acid transcription reaction is oligoribonucleosides having no phosphate group at the 5 ′ end and 5 ′ At least one labeled nucleic acid transcription initiator consisting of oligoribonucleotides having a mono- or diphosphate group at the end is used.
