abstract | (57) [Problem] To provide a transformation system in which a selection marker is eliminated after inserting a target gene into a yeast chromosome. SOLUTION: A DNA comprising a pair of R recognition sequences arranged in the same direction, sandwiching an R gene arranged under the control of an inducible promoter and an expressible selectable marker gene. In a DNA construct in which a DNA fragment capable of recombination with the chromosomal DNA of yeast is directly or indirectly linked to both ends of the fragment, the R recognition sequence is asymmetrically shortened. After expression results in recombination of the R recognition sequence and elimination of the selectable marker, a functional R A DNA construct in which no recognition sequence remains. Since no R-recognition sequence that can function after exclusion of the selectable marker remains, recombination does not occur, and a foreign gene can be inserted multiple times using the same selectable marker. |
