abstract | (57) [Summary]nDisclosed are methods for synthesizing stereospecific (Rp) phosphorothioate oligonucleotides. In this method, a primer comprising a plurality of deoxyribonucleotides and one ribonucleotide at the 5 'end or at the second position from the 5' end is annealed to the template. This construct is contacted with a mixture of deoxynucleoside α-triphosphate Sp diastereomer and DNA polymerase to form a PS-Rp oligodeoxynucleotide extension, which is cleaved after the ribonucleotide in the primer. The oligodeoxynucleotide is released as a single-stranded PS-Rp oligonucleotide. Also disclosed are PS-Rp oligonucleotides and oligonucleotides prepared according to the methods of the invention. |
