abstract | (57) [Summary]nThe present invention relates to plant thioesterases, especially plant acyl-ACP thioesterases having significant activity on stearoyl-ACP. Described are DNA constructs useful for expressing plant stearoyl-ACP thioesterase in plant seed cells. Such a construct provides a control of regulatory elements that, when expressed in a transgenic plant, can preferentially direct the expression of plant stearoyl-ACP thioesterase in seed tissue compared to other plant tissues. Below contains the DNA sequence encoding the desired plant stearoyl-ACP thioesterase. The present invention also relates to a method of modifying stearic acid produced in plant seed cells using a DNA sequence encoding a plant stearoyl-ACP thioesterase. The plant thioesterases with significant activity against stearoyl-ACP exemplified herein are derived from mangosteen. An altered stearic acid concentration can be obtained by expressing mangosteen thioesterase alone or in combination with an antisense construct that reduces natural stearoyl-ACP desaturase in plant seeds. |
