abstract | (57) Abstract The present invention provides recombinant single-chain antibodies that specifically bind to Lewis Y- related homogeneous antigens and fusion proteins comprising those antibodies. More specifically, the present invention provides a single-chain Fv (scFv) region of monoclonal antibodies B1, B3 and B5, a humanized single-chain Fv region of B1, B3 and B5, and a fusion protein comprising those scFvs. provide. The present invention also provides for a number of stabilizing mutations in Lewis Y binding monoclonal antibody B3. In addition, the present invention provides a method for detecting cells having Lewis Y antigen in a patient and a method for killing or suppressing the proliferation of cells having Lewis Y antigen in a patient. The present invention also provides a method of improving the binding affinity of an antibody lacking serine at position 95 of the VH region, comprising changing position 95 to serine. |
