The periodic optical pattern formed from the illumination system (508) is detected by the computer system (540). The computer (540) forms a highly transparent mask (717) for the image (712) of the biological specimen (20). The edges of the image are detected (716) and the low transmittance portions (718) are processed. The horizontal stripe is enhanced (720). The final threshold value determination (722) generates a binary image (727) of the pattern. A stripe in the pattern is detected (724), and a stripe flag (706) and a stripe area (708) are calculated.
