abstract | (57) [Summary]nThe present invention relates to compositions and methods for preventing pneumococcal infection. In particular, the present invention relates to the identification of minimal receptor targets of pneumococcal adhesion to lung and vascular endothelium and to compositions and methods for preventing such adhesion. In particular, the present invention relates to the use of one or more of the following motifs effective for inducing elution of adherent S. pneumoniae from host cells: disaccharide N-acetyl-D-galactosamine β1-3Gal motif, disaccharide N -Acetyl-D-galactosamine β1-4Gal motif and the ability of one or more carbohydrate factors having an N-acetyl-D-glucosamine motif. In particular, compositions containing all three motifs can elute about 100% of pneumococci from lung epithelial cells and venous endothelial cells. In a specific embodiment, the pharmaceutical composition of the present invention is used for treating a pneumococcal infection in which the host cell is a lung epithelial cell. The pharmaceutical composition for use in inhibiting adhesion to lung epithelial cells or eluting adherent bacteria from lung epithelial cells is an aerosol. The present invention further provides formulations for parenteral administration in the treatment of systemic hematogenous infections by preventing or eluting binding to venous endothelial cells. The invention further relates to methods and kits for determining the presence of pneumococcus in a biological sample from a patient. |
