abstract | (57) [Summary]nWhen electrophoresed on a 4-20% gradient acrylamide gel in 25 mM Tris / 192 mM glycine buffer at pH 8.5 in the presence of about 0.1% (w / v) SDS, about 8. Chondroitinase II, a protein with an isoelectric point of 4-8.45 and an apparent molecular mass of 112 kDa, was isolated and purified. Also provided is a method of co-purifying chondroitinase I and chondroitinase II proteins produced by Proteus vargaris by affinity chromatography. The protein can be further purified by metal chelate chromatography. Also disclosed are therapeutic or surgical compositions of isolated chondroitinase I and II or a co-purified mixture of chondroitinase I and II. These compositions are used in a method that selectively or completely removes the vitreous from the neural retina of the eye. |
