(57) [Summary]nThe present invention relates to a construct comprising a) a targeting sequence, b) a regulatory sequence, c) an exon, and d) an unpaired splice donor. The invention further relates to a method for producing a protein in vitro or in vivo, comprising homologous recombination of a construct as described above in a cell. The homologous recombinant cells are then maintained under conditions that permit transcription and translation, and the protein is expressed. The invention further provides for homologous recombinant cells such as primary, secondary or immortalized vertebrate cells, methods for producing such cells, methods for homologous recombination for the production of fusion genes, and alterations in gene expression in cells.
