titleOfInvention Bacteria detection method
abstract (57) [Summary] (with correction) [PROBLEMS] To provide a method for speeding up a sampling step and detection of an amplified DNA fragment after PCR. SOLUTION: Samples such as foods and drinks are homogenized by adding water (in the figure), mixed with magnetic particles coated with an anti-O157 lipopolysaccharide antibody, and the E. coli O157 strain in the sample is collected ( In the figure). A sample obtained by repeating the above operation is placed in a PCR reaction container (in the figure), magnetic particles are attracted from the outside of the reaction container, the magnetic particles are collected, suspended in sterilized water, washed, and then transferred to the PCR reaction container again. Then, a reaction solution for gene amplification (PCR reaction solution) containing FITC-dUTP is put therein, and a PCR reaction is carried out (in the figure). If E. coli O157 strain containing the verotoxin gene is contained in the sample, FITC-dUTP is DNA Since the degree of fluorescence polarization is increased by the incorporation into the chain, Escherichia coli O. The presence of 157 strains can be detected.
