titleOfInvention | How to select highly active ribozymes in vivo |
abstract | (57) Abstract: [PROBLEMS] To provide a method for preferentially selecting a highly active and stable ribozyme in vivo from a group of ribozyme mutants that are actively mutated or modified. SOLUTION: (A) a promoter sequence in order from the upstream side; (C) SD sequence; (D) initiation codon for translation into protein followed by frameshift sequence; (E) transcript of which is a desired sequence cleaved by ribozyme; and (F) dihydrofolate reductase. DNA sequence to A DNA sequence that is thought to contain an active ribozyme gene; is inserted between the sequences (A) and (C) of the "ribozyme selection vector" containing After introduction, the cells are cultured in a medium containing an inhibitor of dihydrofolate reductase, and the active ribozyme gene is taken from the cells that can proliferate. |
