Preparation and use of immune complexes |
abstract |
(57) [Summary]nThe present invention relates to an immunoconjugate comprising an antibody fragment, which antibody fragment is covalently bound to a diagnostic or therapeutic moiety via a carbohydrate moiety near position 18 of the light chain variable region. The present invention is also an immune complex containing an antibody portion, wherein the antibody portion is a complete antibody having a sugar chain forming site near the 18th position of the light chain variable domain, and the sugar chain forming site forms a light chain. It also relates to immune complexes that have been introduced into the antibody by mutating the encoding nucleotide sequence. The resulting immunoconjugate retains the immunoreactivity of the antibody fragment or the complete antibody and targets and delivers the diagnostic or therapeutic component to the labeled tissue where the diagnostic or therapeutic effect is achieved. Thus, the present invention contemplates the use of the above immunoconjugates for diagnosis or immunotherapy. Furthermore, the present invention also relates to a method for preparing the above immune complex. |
