abstract | (57) Summary The present invention provides a selective fusion gene comprising a dominant positive selection gene in reading frame with a negative selection gene and fused to a negative selection gene. The selection fusion gene encodes a single bifunctional fusion protein capable of conferring a dominant positive and negative selection phenotype on the host cell. The dominant negative selection phenotype is conferred by the cytosine deaminase (CD) gene for 5-fluorocytosine sensitivity (5-FC s ). A dominant positive selection phenotype is conferred by, for example, the neo gene for G-418 aminoglycoside antibiotic resistance (G-418 r ) or the hph gene for hygromycin B resistance (Hm r ). The present invention further provides a recombinant expression vector, such as a retroviral vector, containing a selective fusion gene, and a cell into which the recombinant expression vector has been introduced. The selectable fusion genes of the present invention are expressed and regulated as a single genetic material, allowing for highly efficient co-regulation and co-expression. |
