abstract | (57) [Summary]nThe present invention provides methods and compositions for the focused synthesis of branched polymers useful as molecular probes. The present invention also includes some novel branched polymer structures that are particularly useful for detecting target polynucleotides. The branched polymers of the present invention consist of at least two branched chains: at least one branched chain is a target binding moiety capable of specifically binding to the target molecule of interest, and one or more branched chains. Is a signal generating moiety capable of directly or indirectly generating a detectable signal. According to the method of the present invention, branched polymers are combined from components having phosphorothioate groups and / or haloacyl- or haloalkylamino groups. Upon contact, the phosphorothioate group and the haloacyl- or haloalkylamino group react rapidly and efficiently to form a thiophosphorylacyl- or thiophosphorylalkylamino bridge that completes the assembly of the branched polymer. The method of the present invention allows for complete purification and isolation of the components before final combination. The incompletely branched polymer can be easily separated from the desired product by standard means. The method of the present invention allows for the synthesis of several novel branched polymer configurations. |
