abstract | (57) Abstract: Human protein S is a vitamin K-dependent plasma glycoprotein contained in human plasma as a free form and a C4bp-bound form at a low concentration of about 10 mg / liter. Research has revealed that it is involved not only in the role of human plasma coagulation control system, but also in complement reaction and inflammatory reaction, and it has come to be regarded as important in medicine and pharmaceutical fields. Several methods have been proposed so far for separating and purifying this human protein S from human blood, but there has been no method for obtaining a highly purified target product in high yield on an industrial scale. The present inventors have obtained a monoclonal antibody that recognizes free and C4bp-bound human protein S only in the presence of a divalent metal ion, and humanized this on an adsorbent immobilized on an inert carrier. -By contacting a protein S-containing substance in the presence of a divalent metal ion, and then eluting human protein S with a solution containing a metal chelating agent, a high yield, high yield can be achieved without denaturing the target substance. Succeeded in obtaining with purity. |
