titleOfInvention | Yeast mannose-1-phosphate transferase gene and method for producing mannose-1-phosphate-containing acidic sugar chain and phosphate-containing acidic sugar chain using the same |
abstract | (57) Abstract: Yeast (Saccharomyces cerevisiae) mannose-1-phosphate transferase gene, yeast cells transformed with plasmid DNA into which the gene has been introduced are cultured in a medium, A method for producing a mannose-1-phosphate-containing acidic sugar chain, which comprises obtaining a mannose-1-phosphate transferase and allowing it to act on a neutral core sugar chain in vivo or in vitro, and the mannose- 1- A method for producing a phosphoric acid-containing acidic sugar chain, which comprises removing a mannose portion by acid-treating a phosphoric acid-containing acidic sugar chain. [Effects] According to the present invention, an acidic sugar chain obtained by adding mannose-1-phosphate to the same neutral core sugar chain as that of high-mannose produced by mammalian cells such as human is produced by a genetic engineering method using yeast. A large amount and high purity can be produced. Furthermore, to produce a large amount of a useful acidic sugar chain identical to the human-derived phosphate-containing core sugar chain, which can function as a marker for glycoprotein transport to lysosomes in mammalian cells such as humans, in high purity.
