abstract | (57) [Summary] [Purpose] To easily and efficiently impart root-knot nematode resistance to industrially useful cultivars that do not exhibit root-knot nematode resistance traits. [Structure] A method for identifying the presence or absence of root-knot nematode resistance of tomatoes by the genotype of the acid phosphatase-1 gene carried by the root-knot nematodes, wherein the acidity of root-knot nematode-sensitive tomatoes having the nucleotide sequence shown in SEQ ID NO: 1 is used. Phosphatase- Amplified fragment by polymerase chain reaction of 1+ gene (Aps-1 + ) region or acid phosphatase- 11 gene (Aps- 11 ) region of root-knot nematode-resistant tomato having the nucleotide sequence shown in SEQ ID NO: 2. Alternatively, the genotype of the acid phosphatase-1 gene is identified by the length of the fragment obtained by reacting this amplified fragment with a restriction enzyme, and the presence or absence of root-knot nematode resistance of tomato is discriminated by this. |
