abstract | (57) [Summary]nProvided is a nucleic acid hybridization assay method for an RNA target using an RNA binary probe and a ribozyme ligase which is a strict RNA ligase for RNA. A preferred assay involves exponential amplification of signal production. Tetrahymena ribozyme ligase is the preferred ligase for use in the present invention. It may be tethered to constrain it near the junction of the ligation reaction. One assay method according to the present invention is the "tethered ligase chain reaction". Also provided is a kit for performing the assay method according to the present invention. |
