(57) [Summary]nThe present invention provides an isolated DNA molecule comprising a DNA segment encoding the glucagon receptor. And a DNA construct comprising a first DNA segment encoding a glucagon receptor operably linked to an additional DNA segment required for expression of the first DNA segment, and Also provided are host cells containing the various DNA constructs. The present invention also provides a method of detecting the presence of a glucagon antagonist, comprising: (a) in the presence of a glucagon agonist, allowing the compound to bind for a sufficient period of time and conditions to allow binding of the compound to its receptor. Exposure to a recombinant glucagon receptor bound to the response pathway and associated responses through that pathway; and (b) to the glucagon receptor upon stimulation of the response pathway by the glucagon agonist alone. Also provided is a method comprising the steps of detecting a decrease in the stimulation of the response pathway resulting from the binding of a compound, and then detecting the presence of a glucagon anti-inflammatory substance. |
