abstract | (57) [Summary] [Structure] At least the following four reaction steps Step 1: A step of producing inorganic phosphorus in a nucleic acid in a sample by a phosphatase reaction, step 2: a deglycosylated purine derivative formed from the used nucleoside by subjecting the produced inorganic phosphorus to a nucleoside phosphorylase reaction in the presence of an excessive amount of nucleoside Alternatively, a deglycosylated pyrimidine derivative and a phosphorylated substrate are produced, and step 3: the produced phosphorylated substrate is made to produce inorganic phosphorus by the same phosphatase to cause the reaction of step 2 to carry out the reaction of step 2 and step 3 as a cycling reaction. Step 4: A method for quantifying a nucleic acid or a composition for quantifying, which comprises a step of enzymatically quantifying the amount of the deglycated purine derivative or the deglycated pyrimidine derivative produced in the whole process. [Effect] Compared with conventional nucleic acid quantification, it is possible to construct an enzymatic cycling reaction very easily and quantify nucleic acid with high sensitivity and reproducibility. |
