abstract |
(57) [Summary] [Object] To provide a method for measuring carbon dioxide using a novel phosphoenolpyruvate carboxylase which does not require an expensive activator and a reagent therefor. [Structure] In the presence of phosphopyruvate, bicarbonate ions in the sample were reacted with a recombinant phosphoenolpyruvate carboxylase produced using a gene derived from Acetobacter hanseny IFO14820, and oxaloacetate produced was treated with NADH. A method for measuring carbon dioxide gas in a sample by reacting malate dehydrogenase in the presence thereof to measure decreasing NADH, and a reagent therefor. [Effect] As compared with the conventional phosphoenolpyruvate carboxylase derived from acetic acid bacterium, a reagent for measuring carbon dioxide having excellent stability is obtained. |
