abstract | (57) [Abstract] [Object] The present invention relates to a rapid and simple method and apparatus for measuring ammonia using an enzymatic reaction. [Structure] The sample solution is allowed to act on L-glutamate dehydrogenase 4 together with α-ketoglutaric acid and nicotinamide adenine dinucleotide reduced form, thereby consuming the nicotinamide adenine dinucleotide reduced form, and then with L-glutamic acid dehydrogenase. L-lactic acid was produced in an amount corresponding to the reduced form of nicotinamide adenine dinucleotide remaining unconsumed in the reaction of acting on lactate dehydrogenase 5, and the produced L-lactic acid was further converted to L-lactate oxidase 6 A method for measuring ammonia by acting on the above, and detecting an electrode active substance that increases or decreases when L-lactic acid is oxidized by L-lactic acid oxidase. |
