[Structure] Presence of an enzyme source, divalent iron ion and diketoglutaric acid, which is derived from the genus Dactylosporandium or the genus Amycolatopsis and catalyzes the hydroxylation reaction of L-4-proline to trans-4-hydroxy-L-proline Under an aqueous medium, L-4-proline was added to trans-4-hydroxy- A method for producing trans-4-hydroxy-L-proline, which comprises converting to L-proline and collecting the produced trans-4-hydroxy-L-proline from the aqueous medium, and useful for the production method. A method of analyzing a sample containing L-proline 4-position hydroxylase and prolines by high performance liquid chromatography.
