(57) [Summary] [Purpose] A protein that binds calcium ion in a vitamin K-dependent manner from a non-vitamin K-dependent associated protein to a solution without requiring removal of calcium ion and without the accompanying possible denaturation of the protein. To separate. [Structure] Isolation and purification of vitamin K-dependent protein are carried out by utilizing at least anion exchange chromatography and optionally affinity chromatography. [Effect] This method is particularly suitable for purification of not only Factors II, VII, IX, and X, but also Protein S, Protein C, and Protein Z. In this way vitamin K-dependent proteins are obtained with a purity of 95%. |
