abstract |
(57) Summary [Objective] To provide a method of glycosylating an acceptor at a mannosyl group while recycling guanosine diphosphate using mannosyl-1-phosphate in a single vessel. [Structure] In an aqueous medium in a single container, the following (i) The components of (v) are mixed to form an aqueous reaction medium, the aqueous reaction medium having a pH of about 5 to about 10 and a temperature of about 0 to about 40 for a time sufficient for the receptor to be glycosylated. Keep at ℃. (I) mannosyl-1-phosphate; (ii) GDP-mannosyl pyrophosphorylase; (iii) mannosyl transferase; (iv) receptor; and (v) (a) GDP, GTP or both, (b) phosphate. A guanosine diphosphate cycle system comprising a donor, and (c) a kinase that transfers the phosphate group from the phosphate donor to GDP to form GTP. ((Ii), (Iii) and (v) are present in catalytic amounts. ) |
