abstract (57) [Summary] [Object] The present invention relates to a method for decontaminating an amplicon using a single-strand-specific exonuclease. The method of the invention comprises mixing a nucleic acid preparation containing target nucleic acid, potentially contaminated with a nucleic acid amplification reaction product (amplicon), with a single-strand-specific exonuclease and digesting the amplicon. Incubating the preparation with an exonuclease for a time sufficient to The amplicon may be the product of a strand displacement amplification reaction (SDA). Suitable exonucleases include exonuclease I, exonuclease VII, and DNA polymerases having 3'-5 'exonuclease activity.
