abstract (57) [Summary] [Object] To provide a leukocyte measurement reagent containing a hemolytic agent for hemolyzing red blood cells and a method for preparing a leukocyte measurement sample. [Structure] (a) A red blood cell hemolyzing agent comprising a hypotonic solution containing a buffering agent for keeping pH in an acidic range; and (b) A reagent for measuring leukocytes, which comprises a solution comprising a buffer agent for neutralizing the acid in the above buffer agent and an osmotic pressure compensating agent for adjusting the solution to an osmotic pressure that maintains the morphology of leukocytes; and (a) a step of adding fresh blood that has been subjected to anticoagulation treatment to a red blood cell hemolyzing agent comprising a hypotonic solution containing a buffering agent for keeping the pH in an acidic range to hemolyze red blood cells; and (b) the buffering agent A solution comprising a buffering agent for neutralizing the acid in the solution and an osmotic pressure compensating agent for adjusting the solution to an osmotic pressure that maintains the morphology of leukocytes was obtained in the above (a), A method for preparing a sample for measuring leukocytes, which comprises a step of adding to a blood sample treated with an erythrocyte hemolytic agent.
