abstract | (57) [Summary] [Purpose] To improve the problem of impurity associated with nucleic acid amplification reaction. [Structure] A method for improving the impurity problem associated with a nucleic acid amplification reaction using a novel primer or polynucleotide containing a ribose residue that can be separated from an amplification product. Removal of the primer from the product by selective hydrolysis of the ribonucleotide bond prevents it from further amplification. Since the method of the present invention does not change the DNA synthesized by primer extension or the DNA inside the sample in any sense, it can be performed either before or after amplification, or both. The method of the invention can be used in primer-dependent amplification reactions. An example of these amplification reactions is P It is an amplification reaction in which a polynucleotide substrate such as CR, a transcription system or a ligase chain reaction is directly incorporated into a product. The method of the present invention also provides an effective means of separating amplified nucleic acid sequences which consists of first binding the product to a solid support and then removing by splitting the primer. |
