abstract (57) [Abstract] [Purpose] To provide a method capable of conveniently and rapidly measuring the cell activity and the number of viable cells of a large number of samples by using an ordinary microplate reader. [Structure] Oxidized quinone such as 1,4-naphthoquinone, 2-methyl-1,4-naphthoquinone (menadione) or tetramethyl-p-benzoquinone is allowed to act on a measurement system containing living cells to generate hydrogen peroxide. The reaction is performed with an indirect coloring reagent or a direct coloring reagent via an enzyme having hydrogen peroxide as an acceptor, and the color development intensity is quantified with a spectrophotometer (colorimeter). The preferred enzyme is peroxidase, the indirect color reagent is N- (carboxymethylaminocarbonyl) -4,4'-bis (dimethylamino) -diphenylaminosodium) or leuco crystal violet, and the direct color reagent is cytochrome c peroxidase. ..
